When a reply is labeled Unsubscribe, Master Inbox unsubscribes the lead in Smartlead and adds a Do not contact label, so no further Smartlead step goes out.

Set it up in Master Inbox

In Master Inbox, open Settings, then Workflows, and click Add a workflow.
Name it Unsubscribe opt-outs in Smartlead automatically.
Set the trigger to Add Label and choose Unsubscribe.
Add the action Unsubscribe in Smartlead: The lead's active campaigns.
Add the action Add labels: Do not contact.
Save the workflow and switch it to Active.
