Set it up in Master Inbox

In Master Inbox, open Settings, then Workflows, and click Add a workflow.
Name it Start an instant reply sequence for positive replies that arrive on weekends.
Set the trigger to Add Label and choose Interested or Meeting request.
Add a condition: Day is Saturday or Sunday.
Add the action Add labels: Sequence: weekend follow-up.
Save the workflow and switch it to Active.
